Methodology

Polymerase Chain Reaction (PCR) 

 

This test is not recommended for the evaluation of suspected sexual abuse, other medico-legal indications, or for evaluation of children <10 years of age. In these circumstances, culture is the test of choice.

Specimen Requirements

Submit only 1 of the following specimens:

 

Endocervix
Collect specimen using GeneXpert CT/NG Transport Kit as follows:

  1. Remove excess mucus from the cervix and surrounding area using large-tipped cleaning swab provided in collection kit and discard.
  2. Insert the collection swab into the endoceervical canal. Rotate the swab clockwise for 10-30 seconds in the endocervical canal.
  3. Withdraw swab carefully.
  4. Unscrew the cap from the transport tube and immedictely place the specimen swab into the transport reagent tube.
  5. Identifying the scoreline, break the swab shaft against the side of the tube. Discard the top portion of the swab shaft.
  6. Re-cap the swab transport reagent tube and tighten cap firmly.
  7. Label tube with patient’s full name (first, last, and middle initial), date of birth or medical record number (if available), date and time of collection, location of patient, and type of specimen.

Stability:  60 days, refrigerated or room temp, if collected in GeneXpert transport kit.

 

Note:  

  1. Spermicidal agents and feminine powder sprays may interfere with the assay and should not be                    used prior to collection.
  2. Specimen source is required on request form for processing.

 

Urine

  1. Patient should not have urinated for at least 1 hour prior to specimen collection.
  2. Patient should collect first 15 mL to 20 mL of voided urine (first part of stream-not midstream) into a screw-capped, sterile, plastic, preservative-free specimen collection container.
  3. Label container with patient’s full name (first, last, and middle initial), medical record number (if available), date and time of collection, location of patient, and type of specimen.
  4. Send specimen refrigerated. Maintain sterility and forward promptly.

Stability:  Room Temperature:  Male urine - 3 days   Female urine - 24 hours

                Refrigerated:  Male and Female - 8 days
